AB Dendritic cells (DC) are the major antigen presenting cells for the
induction of T cell mediated immune responses. We have found DC isolated
from peripheral blood by sorting are CD4- and do not allow viral entry
by T cell or macrophage tropic isolates of HIV-1. DC are functionally
and phenotypically heterogeneous and immature blood DC and DC from other
sites express CD4. PCR was used to detect entry of macrophage and T cell
tropic HIV-1 into DC expressing CD4. Thymic DC are CD4+ dendritic cells
that develop with thymocytes from a common precursor. Thymic DC were
purified to near homogeneity and pulsed with different HIV-1 isolates.
After 24-48 hrs provirus was detected for macrophage tropic Ba-L and
YU-2 virus strains, but not T cell tropic isolates 228200 or IIIB. In
addition to the previously described ability of mature DC to carry virus
and infect T cells during antigen specific responses, infected CD4+ DCs
may be a continuing source of virus or provide signals for induction of
energy or apoptosis of responding T cells.
